TND1128 is 10-ethyl-3-methylpyrimido[4,5-b]quinoline-2,4-dione<\/strong>, a 5-deazaflavin derivative<\/strong>. It behaves as an NAD\u207a-like\/self-redox compound and seems to facilitate mitochondrial energy metabolism. The 2024 work suggests that it can participate functionally in the TCA-cycle\/electron-transfer-chain redox system<\/strong>, increasing mitochondrial membrane polarization (\u0394\u03a8m) and facilitating ATP synthesis when neuronal energetic demand rises<\/strong>.<\/p>\n

One particularly interesting observation is its effect on Ca\u00b2\u207a handling<\/strong>. In mouse brain slices, pretreatment reduced both cytoplasmic and mitochondrial Ca\u00b2\u207a increases produced by strong neuronal depolarization. That does not necessarily mean TND1128 blocks voltage-gated calcium channels directly<\/strong>; the researchers interpret this largely in the context of improved mitochondrial energetic handling and calcium homeostasis.<\/p>\n

There is also evidence for longer-term neurotrophic\/plasticity-like effects<\/strong>. In cultured mouse hippocampal neurons, TND1128 increased dendritic and axonal branching and increased the number of excitatory synapses. The authors linked these effects to its NAD\u207a-like redox\/mitochondrial activity rather than direct neurotransmitter-receptor stimulation.<\/p>\n

Biochemical Classification:<\/strong><\/p>\n

TND1128 \u2192 mitochondrial bioenergetic\/redox modulator \u2192 \u2191 \u0394\u03a8m \u2192 enhanced ATP availability \u2192 improved Ca\u00b2\u207a handling \u2192 downstream neuronal growth\/plasticity effects.<\/p>\n

Mitochondrial membrane potential: probably the best-established action<\/h2>\n

This is presently the cleanest pharmacological signal.<\/p>\n

Mouse brain slices were obtained after systemic TND1128 treatment. At 10 mg\/kg i.p. 24 hours beforehand<\/strong>, mitochondrial membrane potential was significantly more polarized.<\/p>\n

When neurons were subsequently challenged with elevated extracellular K\u207a, thereby increasing neuronal activity and energetic requirements, TND1128-treated tissue showed a larger demand-associated \u0394\u03a8m response that the investigators interpreted as increased capacity for ATP production.<\/p>\n

This is important because it doesn’t look simply like:<\/p>\n

TND1128 \u2192 indiscriminately forces ATP production continuously.<\/strong><\/p>\n

The authors instead describe something closer to:<\/p>\n

TND1128 \u2192 increases energetic reserve \u2192 ATP synthesis rises when neurons need it.<\/strong><\/p>\n

Hence their phrase “on-demand ATP synthesis.<\/strong><\/p>\n
