TND1128 is 10-ethyl-3-methylpyrimido[4,5-b]quinoline-2,4-dione, a 5-deazaflavin derivative. It behaves as an NAD⁺-like/self-redox compound and seems to facilitate mitochondrial energy metabolism. The 2024 work suggests that it can participate functionally in the TCA-cycle/electron-transfer-chain redox system, increasing mitochondrial membrane polarization (ΔΨm) and facilitating ATP synthesis when neuronal energetic demand rises.

One particularly interesting observation is its effect on Ca²⁺ handling. In mouse brain slices, pretreatment reduced both cytoplasmic and mitochondrial Ca²⁺ increases produced by strong neuronal depolarization. That does not necessarily mean TND1128 blocks voltage-gated calcium channels directly; the researchers interpret this largely in the context of improved mitochondrial energetic handling and calcium homeostasis.
There is also evidence for longer-term neurotrophic/plasticity-like effects. In cultured mouse hippocampal neurons, TND1128 increased dendritic and axonal branching and increased the number of excitatory synapses. The authors linked these effects to its NAD⁺-like redox/mitochondrial activity rather than direct neurotransmitter-receptor stimulation.
Biochemical Classification:
TND1128 → mitochondrial bioenergetic/redox modulator → ↑ ΔΨm → enhanced ATP availability → improved Ca²⁺ handling → downstream neuronal growth/plasticity effects.
Mitochondrial membrane potential: probably the best-established action
This is presently the cleanest pharmacological signal.
Mouse brain slices were obtained after systemic TND1128 treatment. At 10 mg/kg i.p. 24 hours beforehand, mitochondrial membrane potential was significantly more polarized.
When neurons were subsequently challenged with elevated extracellular K⁺, thereby increasing neuronal activity and energetic requirements, TND1128-treated tissue showed a larger demand-associated ΔΨm response that the investigators interpreted as increased capacity for ATP production.
This is important because it doesn’t look simply like:
TND1128 → indiscriminately forces ATP production continuously.
The authors instead describe something closer to:
TND1128 → increases energetic reserve → ATP synthesis rises when neurons need it.
Hence their phrase “on-demand ATP synthesis.
