DSIP Peptide

DSIP (Delta Sleep-Inducing Peptide), also known as emideltide, is a nine-amino-acid peptide with a distinctive place in sleep research. Its appeal centres on the connection between easier sleep onset, more time asleep and better daytime alertness—the possibility of a more settled night carrying through into a more alert day.

✅ Primary Research Profile: Sleep Onset & Sleep Efficiency

• Sleep onset means how long it takes to fall asleep. Sleep efficiency is the proportion of time in bed actually spent asleep. Both are central to DSIP’s sleep-related appeal.
• Early human work reported improvements in these measures. A small study involving 14 people with chronic insomnia also reported improved daytime alertness and performance.

Peptide Signals Involved in Sleep:

• Animal and laboratory studies suggest that DSIP can influence the body’s own opioid signalling system, which helps regulate functions including pain and sleep. In rat brain tissue, DSIP stimulated the release of enkephalins—small messenger peptides involved in that system.
• This offers one possible explanation for some experimental effects; the same study did not find direct binding to opioid receptors. It is not a confirmed human mechanism.

Stress-Related Hormone Signals:

• DSIP has also been studied in connection with ACTH, a messenger that tells the adrenal glands to produce cortisol. One small human study found reduced ACTH-related activity, while cortisol remained unchanged.
• Later experiments did not confirm suppression of ACTH or cortisol, so this remains an area of interest rather than an established stress-relief benefit.
